- Peter Parker/Spiderman: Peter Parker is a 15-year-old boy who was bitten by a genetically altered spider. Peter donned a spider costume and sought to fight crime after allowing a burglar to escape, only to have that same criminal - later identified in this series as Walter Hardy - later kill his Uncle Ben. He learned that "with great power comes great responsibility", a phrase that now drives him to act as a super hero. He is shown to be a cocky, impulsive boy, who wants to save the world.
- Gwen Stacy: Gwen is the daughter of the scientist George Stacy, and is Peter's girlfriend. She appears as a young-looking shy 14-year-old girl. She has ginger hair that appears to be tied up on a small ponytail and curved bangs that go to the right. She also has a yellow headband. April wears an elbow-length black shirt with a pink and white sport shirt with the number "4" over it, white skirt, white high socks with two blue stripes on them, with white high-heeled boots.
- Harry Osborn: Harry is Peter's friend and is the son of industrialist Norman Osborn. He constantly lives in his father's shadow and considers Peter a good friend, though at times also resents him for earning Norman's respect, which Harry has never been able to do.
